from what i have gathered there are a lot of (drop in/interchangeable) engine and trans combos for my 78 k10. my question is what automatic trans can I replace my 4 speed manual with to mate up with the 250 inline 6 in my truck. also is this a simple project to take on myself? the guys I have talked to tell me its a pretty straight forward job. thanks

well all the small block and big block and 6 cyl bellhousings are the same bolt pattern, as for a direct bolt in not sure. you have spline count and length to look at as well as the transfer case adapter.

Im not sure what other changes are necessary but I know a TH350 will mate up, that is what the L6 came factory with
You'll be best off finding a tranny/t-case combo so you know it'll mate up. The TH400/4l80e and TH350/700r4 have completely different output shafts, and your current t-case might not bolt to either (??)...
You should probably look for an auto steering column, as well.
You'll almost definitely need a new driveshaft (or shortened/lengthened, just beware factory shafts are nearly impossible to balance after hacking them for a length change)
you need a flexplate that will mate with whatever transmission you choose. Buy a new one, and use locktite (they're cheap if you shop around). Don't go with a used flexplate unless it's free&good. You'll kick yourself later if a junkyard flexplate breaks because of a hairline crack you couldn't see.
You won't be able to (easily) use a 2wd tranny, because the output shaft is wrong and it won't have the 4wd tail housing with the t-case bolt pattern.

If all you can find is a 2wd trans you could run a divorced T-case

it you want the best of both worlds, but a 700R4/208, and buy the constant pressure valve body. you should be able to get it all for under $500 or so depending on location, including the new valve body.
